The extension's side panel and compose helper call this URL. Make sure you're signed in to the same domain in your browser so the session cookie applies.
When this is on, Flow will append the email signature you've set on Flow's settings page to every generated draft. Leave off if you already have a signature configured in Gmail — the extension will preserve it automatically when inserting the draft.
